Tartrate-resistant acid phosphatase (TRAP, ACP5) is a metalloenzyme belonging to the acid phosphatase family, which catalyzes the conversion of orthophosphate monoesters into alcohols and orthophosphates. It is an iron-containing glycoprotein synthesized as a 35kd monomeric proenzyme. ACP5 is involved in bone resorption and is secreted by active osteoclasts. It dephosphorylates osteopontin (a bone matrix protein), thereby promoting osteoclast detachment and migration. The expression of ACP5 is associated with tumor progression in various cancers such as colorectal cancer, ovarian cancer, breast cancer, and melanoma. In vitro experiments have shown that downregulation of ACP5 expression in colorectal cancer cell lines promotes tumor cell proliferation and invasion. In serum, ACP5 has been used as a marker for thyroid metastasis. In addition, ACPs remove the mannose-6-phosphate recognition markers from lysosomal proteins.
