This ELISA kit uses the Competitive-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with IgG1. During the reaction, IgG1 in samples or Standard competes with a fixed amount of IgG1 on the solid phase supporter for sites on the Biotinylated Detection Ab specific to IgG1. Excess conjugate and unbound sample or standard are washed from the plate, and Avidin conjugated to Horseradish Peroxidase(HRP) are added to each microplate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of stop solution and the color change is measured spectrophotometrically at a wavelength of 450±2 nm. The concentration of IgG1 in the samples is then determined by comparing the OD of the samples to the standard curve.
Rat IgG1 is a subclass of immunoglobulins (IgGs), which are large molecular proteins produced by the immune system. Structurally, IgG1 consists of four polypeptide chains: two heavy chains and two light chains, forming a Y-shaped molecule. The variable regions of IgG1 confer antigen specificity, while the constant regions mediate effector functions.IgG1 plays a pivotal role in humoral immunity, exhibiting high affinity for antigens and activating complement pathways. It is also involved in antibody-dependent cellular cytotoxicity (ADCC) and opsonization. In disease contexts, alterations in rat IgG1 levels can indicate immune dysfunction. For instance, elevated IgG1 levels may be observed in autoimmune disorders or infections. Moreover, IgG1 interacts with various factors, including Fc receptors on immune cells, which regulate its biological activities. The glycosylation pattern of IgG1 also influences its effector functions and therapeutic potential. In summary, rat IgG1 is a versatile immune molecule with a defined structure and diverse functions, playing a critical role in health and disease.
