1. Seal endogenous biotin. Before using biotinylated reagents, treat with an affinity lectin/biotin blocking kit, especially for tissues such as kidneys, liver, and spleen. 2. Optimize reagent concentration. Through pre-experiments (such as titration experiments), determine the optimal working concentrations of biotinylated antibodies and streptavidin-labeled probes to achieve the best signal-to-noise ratio. 3. Reduce non-specific binding. Dilute all reagents with a blocking buffer containing 1% BSA or 10% normal serum; set up a "no antibody" control to evaluate the background signal of the detection system. 4. Ensure biotin accessibility. When using biotinylated peptide segments, introduce flexible spacer arms (such as PEG, Ahx, or G-S) between the biotin and the peptide segment to avoid steric hindrance. 5. Titrate streptavidin conjugates. One streptavidin molecule can bind up to four biotin molecules, but excessive dosage is the main cause of high background, so titration must be performed. 6. Control experimental conditions. When performing cell surface labeling, biotinylate on ice to prevent reagent endocytosis; use compatible buffers (such as PBS, TBS, etc.).

The main differences between the direct method and the sandwich method in ELISA are as follows: 1. The quantity of antibodies is different. In the direct method ELISA, there is only one enzyme-labeled antibody, while in the sandwich method ELISA, there are two antibodies (the coating antibody + the detection antibody). 2. The antigen binding method is different. In the direct method ELISA, the antigen is directly coated on the solid-phase carrier, while in the sandwich method ELISA, the antigen is sandwiched between the two antibodies. 3. The specificity is different. The specificity of the direct method ELISA is lower (the enzyme-labeled primary antibody may bind to non-specific proteins), while the specificity of the sandwich method ELISA is higher (the two antibodies recognize the antigen, with strong specificity). 4. The sensitivity of the direct method ELISA is lower, while that of the sandwich method ELISA is higher. 5. The main applications are different. The direct method ELISA is used for screening a large number of monoclonal antibodies and detecting small molecule antigens (such as drugs, hormones, etc.). The sandwich method ELISA is used for detecting trace proteins, cytokines, etc. in complex samples.
